Driver chooses the music

I've finally seen the Pilot of Supernatural. I love the brotherliness of Sam and Dean. Just the name calling and the smacking and the utter comfort they take from and have with each other. It's fantastic.
My question, though, and I know there probably isn't an answer (yet) is this:
It seems quite clear that the mom (Mary?) was killed because she stopped the Thing from whatever it was going to do with Sam (my guess is it was going to somehow infect Sam with it's essence or something to create either a human slave or a human host [like make Sam an antiChrist]).
Why was Jess killed? Did the Thing not realize Sam wasn't there (which seems... odd). Had Sam fallen asleep and I missed the indication of the passage of time? Was it trying to warn Sam or, conversely, get Sam back into the game and, thus, make him somehow more exploitable? Or what?
God, when is this show going to come back? I want it now.
